siRNA-mediated depletion of drebrin inhibits differentiation of primary myoblasts. (A) C2C12 cells were transfected with nonsilencing (control) or Dbn1-specific siRNA. After 3 days in differentiation medium, cultures were fixed and triple-stained for myosin heavy chain (MHC; red), drebrin (green) and 4',6-diamidino-2-phenylindole (blue). Bar: 20 μM. (B) Quantification of fusion index in cultures from part (A). Con, control siRNA. Values shown are means ± SD of triplicate determinations. *P < 0.01.
