The segment encoding the mature C-terminal domain following the furin processing site is shown in red. In the targeting construct, this segment was replaced precisely with the corresponding segment taken from Gdf11. The neo-cassette, introduced into intron 2 for selection of ES cells, was removed using a germline-expressed cre recombinase, leaving behind a single LoxP site in intron 2.
